I'm afraid that I don't use the same DAW as you but the answer to your question is yes, Serato Sample should/does trigger when you hit play on your DAW but only when it gets activated by a midi part/event/clip.

I don't mean this in a condescending way, but do you realise that you still have to play the midi parts into your song/timeline like you would with any other vst/virtual instrument and then Serato Sample will get triggered by those midi parts.

It isn't like having an MPC synchronised to your DAW which will start as soon as you press play on your DAW/computer, you have to actually play in some parts so that the midi part/clip triggers Serato Sample at a certain point, like any other midi VST would.
